Intravenous administration of heterodimeric phospholipase A2 HDP-1 disturbs cardiovascular system functions and biochemical composition of blood in anesthesiated rats

Heterodimeric phospholipase A2 HDP-1 from the venom of Nikolsky viper consists of two non-covalently bound subunits – HDP-1P and HDP-1In, the first of which exhibits lipolytic activity, while the second does not. The subunits have close molecular masses and similar amino acid sequences. Administration of HDP-1 and HDP-1P to anesthetized rats is accompanied by prolonged hypotensive effects, the intensity of which depends on the dose. With the administration of HDP-1P, the effect develops more slowly than in the case of HDP-1 containing both subunits, while individual HDP-1In does not affect hemodynamic parameters. It was found using electrocardiography that HDP-1 and HDP-1P affect the functioning of the rat heart, causing arrhythmia, which may indicate their ability to affect the innervation of the heart. The administration of HDP-1 and its subunits also leads to changes in biochemical blood parameters, indicating their damaging effect on cells and internal organs.

New cellular partners of HIV-1 integrase and their role in viral replication

Cellular proteins, partners of viral enzymes, are involved in the replication of the human immunodeficiency virus type I (HIV-1) at various stages. Thus, the viral enzyme integrase, participating in several stages of the viral cycle, interacts with various cellular proteins. A number of them are already known, and for some the mechanism of action has been established, but the search for integrase cellular partners continues. In this work, the identification of cellular partners of HIV-1 integrase has been carried out by the cross-linking method followed by mass spectrometry (XL-MS). Twelve new potential integrase partners have been identified, and some of them have been examined for their effect on the early stages of HIV-1 replication.

The role of aquatic ecosystems as a source of omega-3 polyunsaturated fatty acids for the sand lizard Lacerta agilis (Linnaeus, 1758), inhabiting a shore biotope

The composition and content of fatty acids of the sand lizard (Lacerta agilis) and its potential food objects in the shore and steppe biotopes were studied. For the first time, it was established that the content of long-chain omega-3 polyunsaturated fatty acids, primarily eicosapentaenoic acid (EPA), significantly increases in the tissues of lizards from a shore biotope during the period of emergence of amphibious insects from the nearby salt Lake Shira. Among all the food sources of lizards, the highest EPA content was found in chironomids imago. The content of docosahexaenoic acid (DHA) in the muscles of the lizards was also high, although this acid was not detected in the invertebrates that the lizards consumed. In this regard, it has been suggested that the sand lizard is able to biosynthesize DHA from biochemical precursors contained in food.

Comparative analysis of nutritional and biotechnological value of muscle tissue of mesopelagic fishes of the North-East Atlantic region

A comparative analysis of some nutritional and biotechnological characteristics of muscle tissue of the most widespread and abundant mesopelagic fish species inhabiting the Irminger Sea (North Atlantic) was carried out. At the level of lipid metabolism, potential nutritional value was established for non-commercial Notoscopelus kroyeri and Serrivomer beanii along with the commercial beaked redfish Sebastes mentella. For Lampanyctus macdonaldi and, partially, Borostomias antarcticus, possible biotechnological value was shown in terms of extracting individual valuable molecules and substances of lipid nature. The other studied species (Bathylagus euryops, Chauliodus sloani, Malacosteus niger, Scopelogadus beanii, Stomias boa), as well as B. antarcticus, are very promising for their use in aquaculture in the development of innovative feed compositions and additives for them. It was found that the depth and area of catch don’t have a significant effect on the nutritional and biotechnological characteristics of the muscle tissue of the studied species. The exception is B. euryops, the characteristics under consideration of which vary depending on the area of catch, and for S. mentella and S. beanii – on the depth of catch. The results of the study can serve as a basis for the development and improvement of methods and protocols for the rational use of deep-sea biological resources in the process of their development in the North Atlantic region, and also determine the areas of practical application of these species and the type of products obtained from them or with their use (feed, additive, pure substance, composition, functional food product, etc.).

Intravital optical bioimaging of ovarian cancer using a luminescent cell line

The method of intravital bioimaging based on luminescence occupies an important place in the development and testing of antitumor drugs on model animals and is an essential part of preclinical studies. Bioimaging based on luminescent systems, compared with fluorescent bioimaging, provides a high signal-to-noise ratio, which justifies the development of cell lines, that stably express luciferase genes for subsequent use in model animals. The work describes the creation of a stable cell line SKOV3.ip1-NanoLuc constitutively expressing the NanoLuc luciferase gene. The developed cell line was shown to be effective for intravital luminescence bioimaging of immunodeficient animals with deep-seated intraperitoneal tumors, which can be considered as a model of late-stage ovarian cancer.

Antitumor effect of helium ions with energy of 320 MeV/ion during irradiation of Ehrlich ascites carcinoma cells ex vivo

The regularities of tumor induction and growth were studied in mice exposed to a single ex vivo irradiation with a helium ion beam of Ehrlich adenocarcinoma ascites cells (EAC) at doses of 10 Gy and 20 Gy in two positions of the Bragg curve (before the peak and at the peak) in comparison with X-ray radiation at the same doses. It was shown that the frequency of induction and delay in tumor appearance depend on the dose of helium ion irradiation. The following were determined: the time of a fivefold increase in the EAC volume, tumor growth inhibition, tumor growth index (TGI), and increase in life expectancy (ILE) in mice. A decrease in the TGI values and an increase in the ILE values occurred with increasing dose for all types of radiation. The relative biological effectiveness value for helium ions determined by the area under the EAC growth dynamics curves reached a maximum value of 1.8 upon irradiation at the Bragg peak at a dose of 20 Gy.

Evidence for functionality of transmembrane domains of predicted non-canonical plant phosphotransmitters

Bioinformatic methods have been used to predict a new subclass of proteins among plant phosphotransmitters involved in the signaling system of multistep phosphorelay. In contrast to the canonical soluble nucleocytosolic forms, the found phosphotransmitter sequences, belonging to a wide range of plant taxa, potentially contain transmembrane domains. This suggests localization of such proteins on cell membranes and, therefore, a different function in signaling than that of canonical phosphotransmitters. We tested the functionality of the transmembrane domain of the phosphotransmitter using the protein of the tea plant Camellia sinensis. Membrane localization of the transiently expressed recombinant phosphotransmitter with this domain was confirmed by microscopy and immunoblotting. Thus, for the first time, experimental evidence was obtained for the existence of membrane-bound plant phosphotransmitters with as yet unknown functions. These data suggest the presence of a non-canonical membrane branch of signal transduction in the multistage phosphotransfer system in plants.

The correlation between rhythmic changes of body temperature of small passerine birds (Chloris chloris) and non–tidal gravity fluctuation in the 12–30 minute period range

Previously, a connection between animal activity and deformational changes in the lithosphere was established. We have investigated this phenomenon using the example of the relationship between the body temperature dynamics of males of the common greenfinches (Chloris chloris) and gravimeter data. For a 12-30-minute range of periods, a simultaneous change in the power of the prevailing harmonics of body temperature and gravimeter readings is shown, which indicates the presence of a connection between these processes. It is shown that a stable positive correlation occurs only during the period of bird activity. Therefore, fluctuations in the gravimeter readings in the 1230 minute range of periods can be considered as a marker of an as yet unidentified biotropic environmental factor affecting animal activity.

Finding eggs of the trematode Echinochasmus sp. (Trematoda, Echinochasmidae) in the north of Western Siberia in the late Holocene

A parasitological analysis of 24 dog coprolite samples, 1 cultural deposit, and 4 dung samples obtained during excavations of the Mangazeya settlement (66°42’ N, 82°16’ E), the first Russian city of the 17th century (late Holocene) in the north of Western Siberia was performed. Helminth eggs were found in 26 (89.7%) samples. Echinochasmus sp. eggs were found in 48.3% of the samples: in the cultural deposit, in 2 dung samples, and in 11 dog coprolite ones. This genus is currently absent in the helminth fauna of the region. It could have entered the region with infected dogs, pigs, and humans who arrived to the region from European Russia. The introduction of the helminth did not lead to the formation of a new stable natural focus of echinochasmosis, which is associated with the unfavorable climatic conditions of the region.
